All changes to your running gitolite instance are made on the checked out gitolite-admin repo. They come into effect as soon as you push them to the server.
- Open conf/gitolite.conf in your editor
- Search for all occasions of the users name
- Remove the user completely or adjust permissions
- Commit and push the changes back to the server
